Why should you not shake a vial after reconstituting it?

Updated: 2026-09-09 · Reconstitution and syringes

Short answer

Some protein and peptide formulations are sensitive to vigorous agitation and to the air-liquid interfaces that foam creates, and stability can be affected in those cases. That does not mean there is a universal 'never shake' rule for every product: the correct handling is the one the product itself specifies.

In plain English

  • Foam signals mechanical stress and air-liquid interface in the solution.
  • Sensitivity to agitation depends on the specific formulation, not on 'peptides' in general.
  • The product's own handling instructions are what govern.

What we know

  • Denaturation from mechanical and interfacial stress is well known in protein formulation.

What we don't know yet

  • How much activity each specific peptide loses when shaken is rarely quantified publicly.
