How do I correctly reconstitute a peptide vial?

Reconstituting means adding the sterile diluent that the product specifies to a lyophilized product. Which diluent, what volume, how it is handled, how it is stored and how long it can be used afterwards are product-specific facts. Where an approved label or a pharmacist or clinician instruction exists, that specific instruction is the one to follow; there is no general technique valid for every peptide.

How much bacteriostatic water do I need for a 5 mg vial?

'5 mg' on its own does not let you work out the correct volume. If the product has approved labeling or a pharmacy or clinician instruction, the diluent and the volume are the ones specified there. Separately from that, as pure arithmetic: water does not change how much peptide is in the vial, it only changes the concentration in milligrams per millilitre.

How much bacteriostatic water do I need for a 10 mg vial?

It changes concentration, and with it how precisely you can measure. With little water, being off by a single syringe mark represents much more product. With more water, the same error matters less, but each draw takes more volume. The total peptide in the vial never changes.

A U-100 syringe holds 100 units in 1 ml, so each unit is 0.01 ml. The conversion takes two steps: work out the vial concentration in mg per ml, then divide the amount you want to draw by that concentration to get millilitres, and multiply by 100 to get units.

Can I use sterile water instead of bacteriostatic water?

They are not the same. Sterile water has no preservative: once opened and punctured, nothing holds back bacterial growth. Bacteriostatic water contains benzyl alcohol, which lets the same vial be used several times over a period. That is why bacteriostatic water is the usual choice for multi-dose vials when the product labeling allows it — it is not a default diluent for every product.
